Open the Run box then run the command
chkdsk [drive:] /f
For example: chkdsk c: /f to check and fix errors on c:.
If the drive is locked by the system or some processes, you'll need to tell checkdisk to check the drive on next reboot.

Just a thought ....if your HDD is nearly full, you may hit probs with defrag, as it needs space to move files around, during the process.
It needs at least 15% free space and on some drives, as much as 25%, especially if the drive is small.

Well, your drive has more serious problem(s) than I thought :) such as bad sectors/clusters. This indicates the file system on that volume (drive) is in an inconsistent state due to data corruption or sudden shutdown of the system (causing the dirty bit to be set).
Try this command instead:
chkdsk [drive:] /f /r
The /r switch is to tell chkdsk to locate bad sectors and then recover readable information.

The /r switch of chkdsk I mentioned above will clear the dirty bit set by the system due to inconsistencies found on the HD without the need to switch to safe mode.
